Les deux peuvent parfois être interchangeables pour "penser à quelqu'un/quelque chose" :
I often think of/about you. (Je pense souvent à toi)

THINK OF = opinion, avis ("What do you think of...?") — ou se souvenir de / trouver une idée ("I can't think of his name", "Can you think of a better example?")
THINK ABOUT = réflexion, considération ("I'm thinking about...")
